AgRP neurons: a switch between peripheral carbohydrate and lipid utilization.

چکیده
AgRP/NPY neurons are critical regulators of body weight and food intake. Concordant with their orexigenic effects, it is expected that AgRP ablation leads to the appearance of a lean phenotype. In the current issue of The EMBO Journal, Joly-Amado et al (2012) describe an obese phenotype in a model of AgRP-ablated mice, and link it to a shift in metabolic profile in efferent tissues such as the liver, muscle and pancreas. Hypothalamic AgRP/NPY neurons are known to play a key role in the regulation of body weight and food intake. With the advent of ‘toxin-receptor mediated cell knockout’ technology, several reports have tried to address the physiological relevance of this set of neurons. The ablation of Agrp-expressing neurons has different consequences depending on the age of the mice. Thus, in the neonatal stage, temporal deletion of AgRP neurons by diphtheria toxin

Hypothalamic AgRP-neurons control peripheral substrate utilization and nutrient partitioning.
Obesity-related diseases such as diabetes and dyslipidemia result from metabolic alterations including the defective conversion, storage and utilization of nutrients, but the central mechanisms that regulate this process of nutrient partitioning remain elusive. As positive regulators of feeding behaviour, agouti-related protein (AgRP) producing neurons are indispensible for the hypothalamic int...
متن کاملThe hypothalamic arcuate nucleus and the control of peripheral substrates.
The arcuate nucleus (ARC) of the hypothalamus is particularly regarded as a critical platform that integrates circulating signals of hunger and satiety reflecting energy stores and nutrient availability. Among ARC neurons, pro-opiomelanocortin (POMC) and agouti-related protein and neuropeptide Y (NPY/AgRP neurons) are considered as two opposing branches of the melanocortin signaling pathway. In...
متن کاملRibosomal S6K1 in POMC and AgRP Neurons Regulates Glucose Homeostasis but Not Feeding Behavior in Mice
Hypothalamic ribosomal S6K1 has been suggested as a point of convergence for hormonal and nutrient signals in the regulation of feeding behavior, bodyweight, and glucose metabolism. However, the long-term effects of manipulating hypothalamic S6K1 signaling on energy homeostasis and the cellular mechanisms underlying these roles are unclear. We therefore inactivated S6K1 in pro-opiomelanocortin ...
متن کاملPGC-1α expression in murine AgRP neurons regulates food intake and energy balance
OBJECTIVE Food intake and whole-body energy homeostasis are controlled by agouti-related protein (AgRP) and pro-opiomelanocortin (POMC) neurons located in the arcuate nucleus of the hypothalamus. Key energy sensors, such as the AMP-activated protein kinase (AMPK) or sirtuin 1 (SIRT1), are essential in AgRP and POMC cells to ensure proper energy balance. In peripheral tissues, the transcriptiona...
متن کاملArcuate AgRP neurons and the regulation of energy balance
The arcuate nucleus of the hypothalamus contains at least two populations of neurons that continuously monitor signals reflecting energy status and promote the appropriate behavioral and metabolic responses to changes in energy demand. Activation of neurons making pro-opiomelanocortin (POMC) decreases food intake and increases energy expenditure through activation of G protein-coupled melanocor...
